摘要

胰腺导管腺癌（ pancreatic ductal adenocarcinoma，PDAC）由于常进展到晚期才被诊断并对化疗具有耐药性，因此是一种临床上具有挑战性的癌症。然而，最近对PDAC 的生物学研究进展揭示了早期检测和靶向治疗PDAC 的新机会。本文就PDAC 的发病机制研究进展进行评述，包括肿瘤细胞的分子改变、肿瘤微环境的细胞改变和人群水平的危险因素。本文还分析了PDAC 的早期检测、高危人群的筛查以及治疗方法，可供临床借鉴。

Abstract

Pancreatic ductal adenocarcinoma（PDAC）is a clinically challenging cancer because it is often diagnosed at an advanced stage and resistant to chemotherapy. However，recent advances in biological research on PDAC have revealed new opportunities for early diagnosis and targeted therapy of PDAC. This article reviewed the progress of pathogenesis of PDAC，including molecular changes in tumor cells，cellular changes in tumor microenvironment，and population-level risk factors. The early detection of PDAC， screening and treatment of high-risk population were also discussed for clincal reference.
