Effects of vitamin D3 intervention on blood lipid metabolism in rats with nonalcoholic fatty liver disease[J]. Journal of Southwest Medical University, 2022,45(4):308-314. DOI: 10.3969/j.issn.2096-3351.2022.04.006.
Objective To investigate the effects of vitamin D3 intervention on body weight, blood lipid and liver function in rats with non-alcoholic fatty liver disease. Methods Wistar rats were fed with high-fat diet for 6 weeks to establish nonalcoholic fatty liver model, and then randomly divided into control group (CON group),model group (HFD group),vitamin D3 intervention group (HFD+VD3 group) and solvent control group (HFD+Oil group). After 7 weeks of intervention, serum and liver tissue samples were collected, body weight and liver index were measured, serum biochemical indexes and liver function indexes were determined by automatic biochemical analyzer, and liver histopathological changes were observed by HE staining. Results Compared with the CON group, the serum 25(OH)D3 level decreased significantly (P < 0.05),liver index increased (P < 0.001). Serum levels of TG,TC and LDL-C increased significantly (P < 0.05). HE staining microscopy showed obvious steatosis, ballooning change, small focal necrosis of liver cells with inflammatory cell infiltration. Compared with the HFD+Oil group, the serum 25(OH)D3 level increased significantly (P < 0.05) and the liver index decreased significantly (P < 0.05) after the intervention of vitamin D3. The levels of TG,TC,LDL-C,AST and ALT in serum and the levels of TG and FFA in liver tissue decreased significantly (P < 0.001). The degree of adipose lesion in liver tissue decreased in different degrees. Conclusion Vitamin D3 at 12.5 μg/kg had positive effects on reducing liver index, correcting liver metabolism disorders and repairing endothelial injuries of liver cells.
