OBJECTIVE: To explore the potential mechanism of mulberry leaf-mulberry parasite in the treatment of diabetic nephropathy(DN) based on network pharmacology and bioinformatics. METHODS: Components of mulberry leaf-mulberry parasite and genes of DN were obtained from Traditional Chinese medicine systems pharmacology database and analysis platform
GeneCards and other databases. The differential genes between DN and the healthy people were screened from the GEO database. Cytoscape and R language were used to perform gene ontology(GO) functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG) pathway enrichment analysis of common genes and differential genes. Results of KEGG pf two genes were intersected and verified by molecular docking method. RESULTS: There were 31 active components for mulberry leaf-mulberry parasite
which corresponded to 204 genes
3 566 DN-related genes and 139 common genes. The GSE142153 dataset met the criteria and 113 differential genes were obtained. KEGG pathway enrichment analysis showed that the main involved pathways were advanced glycation end products-receptor for advanced glycation end products signaling pathway in diabetic complications
tumor necrosis factor signaling pathway. Results of molecular docking showed that kaferol and quercetin had good binding activity with cyclooxygenase 2
protein kinase B and cysteine aspartic acid protease. CONCLUSIONS: Mulberry leaf-mulberry parasite can treat DN by giving full play to the role of anti-inflammation and the maintaining of podocyte homeostasis through the multi-target and multi-pathway complex mechanisms.
