OBJECTIVE: To investigate the active components and molecular mechanism of Danning tablets in the treatment of chronic cholecystitis based on network pharmacology. METHODS: The chemical components of Danning tablets were obtained from the TCMSP database and herbal group identification database
and the active ingredients and potential targets were screened according to pharmacokinetic parameters; the targets related to chronic cholecystitis were obtained from the GeneCards
DisGeNET and Drugbank databases
and Venn diagram was drawn with Venny 2.1.1 to obtain the shared targets; the STRING database was used to construct protein-protein interaction network of compounds and shared disease targets
then Cytoscape 3.7.2 software was used to implement visualization and perform network topology analysis to screen the core targets
finally the Cluster Profiler GO. R plug-in was used to conduct enrichment analysis on the core targets. RESULTS: There were 60 shared targets in Danning tablets and chronic cholecystitis
among which the key targets were tumor protein p53
CDKN1 A
CCND1
RAF1
epidermal growth factor receptor
vascular endothelial growth factor A
tumor necrosis factor
matrix metalloproteinase 9
ERBB2 and RB1
etc.
mainly involving bladder cancer
human cytomegalovirus infection and signaling pathways such as phosphatidylinositol-3-kinase/protein kinase B. CONCLUSIONS: The treatment of chronic cholecystitis with Danning tablets may be related to anti-inflammatory and regulation of cholesterol metabolism.
