OBJECTIVE: To investigate the effects of different processing methods on content determination of atropine and scopolamine in different processed products of Datura stramonium L.
so as to ensure the safety and effectiveness of medication. METHODS: The traditional processing methods of glycyrrhiza and vinegar were used to process the raw materials of Datura stramonium L. in Xinjiang. The content changes of atropine and scopolamine were used as the study indexes
the content was determined by high performance liquid chromatography( HPLC). The chromatographic conditions were Super C
18
column( 4. 6 mm × 250 mm
5 μm)
the mobile phase was methanol-water( V∶ V = 30∶ 70
the water contained 0. 02 mol/L sodium acetate solution and 0. 02% triethylamine
p H was adjusted to6. 0 with acetic acid). The flow rate was 1. 0 ml/min
the detection wavelength was 215 nm
the column temperature was room temperature( 25 ℃)
and the sample injection volume was 10 μl. RESULTS: The average content of scopolamine and atropine in Datura stramonium L. by glycyrrhiza was 1. 084 mg/g and 4. 521 mg/g respectively. The content of scopolamine and atropine in Datura stramonium L. by vinegar was 1. 098 mg/g and 3. 293 mg/g respectively. CONCLUSIONS: Through the comprehensive comparison
it is determined that the detoxification effects of Datura stramonium L. by glycyrrhiza is better than that of vinegar.
