Objective:To explore the effects of angelica polysaccharides(APS) on the cell cycle and cell apoptosis rate in bone marrow mononuclear cells(BMNCs) of radiation injured mice
so as to illuminate the molecular mechanism of APS sheltering radiation injured haematogenesis. Methods:The injured mice were divided into 3 groups:normal control group
normal saline group
and APS group.Normal saline group and APS group were given NS or APS for 6 days continuously after the establishment of radiation injured mice model by X ray.Techniques of peripheral blood cells and BMNCs count were made.p53mRNA in murine BMNCs were assayed by RT-PCR.The cell cycle and cell apoptosis in BMNCs were detected by FCM(flow cytometry) on the 7th day. Results:Compare with those in NS group
the numbers of the peripheral leukocytes
erythrocytes
platelets and BMNCs in APS group increased significantly
while the expression of p53mRNA
the percentage of G0/G1 phases
and cell apoptosis rate obviously decreased. Conclusion:APS can decrease the the expression of p53mRNA
improve the chaos of cell cycle
suppress cell apoptosis in radiation injured mice
which can help the recovery of hematopoietic function in mice.
