Objective:To explore the effects of ΔNp63 on invasion in TCCB 5637 cells in vitro. Methods:siRNA were designed according to different targets on ΔNp63 gene were transfected into TCCB 5637 cells by lipidosome method. The TCCB 5637 cells were divided into control group(without transfection)
vector group(without transfection but added lipo2000)
negative plasmid group(with transfection of siRNA negative)and the interference plasmid(with transfection of effective siRNA).The stable colonies were obtained by G418 screening.Laser confocal microscopy was used to detect the location ofΔNp63 in the 5637 cells; The expression level of ΔNp63 was assayed by real-time PCR and Western blot. The invasion of cell was detected by Transwell. The change of cell adhesion was analyzed by cell adhesion assay. Results:The ΔNp63 proteins were localized on the perinuclear region; The expression of ΔNp63 was obviously decreased in the interference plasmid group;in which cell adhesion was decrease as well as the invasion increased significantly.Statistics analysis shows the interference plasmid group has significant difference compared with other groups. Conclusion:The ΔNp63 gene can significantly increased the function of invasion in TCCB 5637 cells.
