Objective: To investigate the effect of RNA interference targeted to Livin on the expressions of apoptotic related proteins Caspase-3 and Caspase-9 in esophageal Eca-109 carcinoma cell. Methods: Transfection of lentiviral vector was employed to perform effective RNA interference targeted to Livin.Fluorescent quantitative PCR and Western blot was used to determine the mRNA and protein levels of Livin
Caspase-3 and Caspase-9 in groups. Results: The expression of mRNA and protein level of Livin in experimental group was significantly lower than that in normal control group and negative control group(P<0.01).The expression of mRNA and protein levels of Caspase-3 and Caspase-9 in experimental group were significantly higher than those in normal control group and negative control group(P<0.01)
respectively. Conclusion: RNA interference targeted to Livin could decrease the expression of Livin effectively.Meanwhile
the mRNA and protein of apoptosis related signal molecules such as Caspase-3 and Caspase-9 were enhanced significantly.
