Objective:To observe the influence of Tanshinone ⅡA on cell proliferation and vascular endothelial growth factor expression in human bile duct carcinoma cell line HCCC-9810
and to explore its possible anti-tumor mechanism of action. Methods:Human bile duct carcinoma HCCC-9810 cells were cultured in vitro and treated with tanshinone ⅡA.The cell proliferation was examined by the method of MTT;the transcriptions of VEGF mRNA were demonstrated by RT-PCR technique;Western blot was applied to examine the protein expression of VEGF.Results:Low-dose(0.1 mg/L)tanshinone ⅡA has no significant inhibitory effect on cellular proliferation of HCCC-9810 cells. When the concentration exceeded 0.25 mg/L
tanshinone ⅡA significantly inhibited cellular proliferation in a time-and concentration-dependent manner
and down regulated the mRNA and protein expression of VEGF(P<0.01).Conclusion:Within a certain drug concentration
tanshinone ⅡA can inhibit the proliferation of HCCC-9810 cells and play a potential role in inhibiting angiogenesis by down-regulating the expression of VEGF.
