Objective:To observe the change in expression level ofp75NTR(p75 neurotrophin receptor)in cultured hippocampal neurons during glutamate-induced excitotoxicity. Methods:Excitotoxicity was induced by glutamate in rat cultured hippocampal neurons; RT-PCR and western blot were used to detect the change in expression level of p75NTR.Results:After glutamate exposure
p75NTR mRNA(P<0.01)and protein(P<0.05)levels had evaluated markedly compared with control group;MK-801
a specific antagonist of N-methyl-D-aspartate recepto(rNMDAR)could decrease p75NTR mRNA and protein levels dramatically compared with glutamate group (P<0.05).Conclusion:p75NTR plays an important role in neuronal damage during glutamate-induced excitotoxicity.
