Objective:To investigate the effects of Angelica polysaccharides on hematopoietic type of acute radiation sickness in mice. Methods:Techniques of peripheral blood cells and BMNC count were made;flow cytometry detected the expression of CD49d
CD44 on Sca-1+BMNC and the cell cycle on BMNC
the proliferation of BMNC were evaluated by MTT assay. Results:Ascompared with those in control group
the numbers of the peripheral leukocyte
erythrocyte
platelet and BMNC in NS (normal saline) group decreased significantly;the expression of CD49d
CD44 on Sca-1+BMNC and the proliferation of BMNC declined;and the percentage of G0/G1 phases significantly increased.APS pretreatment could promote peripheral blood cells and BMNC numbers to recover
decrease the expression of CD49d
CD44 on Sca-1+BMNC in earlier period to make the hematopoietic function of radiation injured mice in pretreated groups to recover more quickly
as compared with APS treated groups receiving the same dose at the same time. The proliferation of BMNC and the percentage of G0/G1 phases in APS pretreated groups had no significant differences from those in APS treated groups receiving the same dose at the same time (P>0.05).Conclusion:The results support our hypothesis that APS can be used to treat the hematopoietic type of acute radiation sickness.APS pretreated groups had better results than APS treated groups in initial observation.
