Objective:To study the effect of gene Kai1 on the proliferation and cell cycle of laryngeal carcinoma Hep-2 cell line. Methods:The Hep-2 cell line was used as the subject
the expansion purification and the concentration of the Kai1 gene was tested
and the gene was transferred into Hep-2 cell line by adenovirus. The non-transferred group was used as a control.By means of MTT the effect of Kai1 gene on the cell proliferation of Hep-2 cell line was tested and by means of RT-PCR the expression of Kai1 in transfected cell was tested;cell cycle of Hep-2 cells was assayed by flow cytometry. Results:The concentration of Kai1 gene can be 1010 PFU/ml after expansion and purification
when the concentration of the adenovirus was 30 MOI
the transfection rate of Hep-2 cell line can be above 93%.MTT shows the cell proliferation was slower and less of the Kai1-Hep-2 cell line compared to the control group(P<0.01).The inhibition rate can up to 29%.Flow cytometry showed the number of Kai1-Hep-2 cells in G0/G1 phase was elevated
and the number in S phase was decreased. Conclusion:The Kai1 gene had the effect of inhibiting the cell proliferation of Hep-2 cell line and the possible machanism would be the cycle pattern was changed after Hep-2 cells was transfected with Kai1 gene.
