摘要

目的:探讨蛋白激酶B（Protein kinase B

PKB/Akt）在肝细胞肝癌（Hepatocellular carcinoma

HCC）组织及癌旁肝组织（Liver tissue just around the tumor

LAT）中的表达变化

并分析其与HCC临床分级之间的关系。方法:采用逆转录聚合酶链反应（Reverse transcription-polymerase chain reaction

RT-PCR）、免疫印迹（Western blot）以及免疫组织化学染色方法检测HCC组织标本中癌及癌旁Akt的表达变化

并分析Akt的表达与HCC临床分级之间的关系。结果:Akt在HCC癌组织中的表达高于癌旁组织

Akt在癌组织中表达的阳性强度与临床分级呈正相关

癌组织中过表达的Akt在细胞核的定位明显增强。结论:Akt的过表达在HCC的发生、发展中具有重要作用。

Abstract

Objective:To investigate the expression of Akt proteins in hepatocellular carcinoma(HCC)and it’s relation with clinical grading.Methods:Firstly

the mRNA levels of Akt in HCC and liver tissue just around the tumor(LAT)were detected by semi-quantitative reverse transcription and the polymerase chain reaction(RT-PCR).Then

analyzing the expression of Akt proteins in HCC and LAT by Western blot and immunohistochemical methods.Finally

the relation between Akt proteins level and HCC clinical grading was analyzed. Results:The expression of Akt in HCC are much higher than LAT

and there is a positive correlation between Akt proteins level and HCC clinical grading.Furthermore

Akt is highly concentrated in the nucleus of HCC.Conclusion:The over-expression of Akt may play an important role in the occurrence and development of HCC.
