摘要

目的:观察XIAP小分子抑制剂Embelin对肿瘤细胞株SGC7901的作用。方法:实验分SGC7901

SGC7901+Embelin两组

采用RT-PCR

Western blot分别测定XIAP mRNA和蛋白水平;MTT法检测SGC7901对VCR

Embelin

VCR+Embelin的敏感性

并计算合用指数CI;体内观察抑瘤情况。结果:体外实验显示Embelin诱导后SGC790的XIAP mRNA基因和蛋白水平均明显高于SGC7901+Embelin组

差异有统计学意义（P

0.05）。MTT显示VCR IC

=0.0458μmol;Embelin IC

=6.3691μmol;联合用药IC

=0.039 9μmol。合用指数CI在抑制率为90%时的抑制率开始大于1。体内实验:VCR组

Embelin组及联合用组分别为重量抑瘤率分别是90.719 2

93.514 4

96.592 3。结论:XIAP小分子抑制剂Embelin对SGC7901有效。联合常规化疗药物VCR作用表现为小剂量协同

大剂量拮抗。

Abstract

Objective:To oberserve the effect of small molecule anatagonists of XIAP（Embelin）on the multidrug resistance cells SGC7901 in vitro and in vivo.Methods:in vitro two groups of cultured SGC7901 cells were used:SG7901（group A）

the SGC7901 cells treated with Embelin（group B）

XIAP mRNA and protein levels of the cells of both groups were determined by TR-PCR

Western blot.MTr assay was performed to examine the IC

and CI.In vivo nude mice were divided into four groups:control group

VCR group

Embelin group

VCR assistance with Embelin group

the weight of tumor growth in the nude mice were measured.Results:Between group A and group B there was a significant difference in the expression of XIPA （P

0.05）

VCR IC

=0.045 8μmole;Embelin IC

=6.3691μmol;Combination IC

=0.039 9μmol

1 at the high inhibition ratio

90% by MTT assay

in vivo experiment tumor weight inhibition rates were93.514 4

96.592 3respectivly. Conclusion:Embelin was good for cells SGC7901

especial for a low dose combination with VCR.
