摘要

目的:探讨大鼠局灶性脑缺血再灌注后脑组织内血管内皮细胞生长因子（VEGF）及VEGFmRNA的表达及意义。方法:制备大鼠局灶性脑缺血再灌注模型

动物分为缺血2h再灌注6h、12h、2 4h、4 8h、72h及假手术对照组。应用免疫组化法检测VEGF蛋白的表达;RT -PCR法检测VEGFmRNA的动态变化。结果:大鼠缺血再灌注6h

缺血侧脑组织梗死灶周边VEGF免疫阳性反应已出现

2 4h明显增多

4 8h达高峰;VEGFmRNA在6h开始上升

12h达高峰

2 4h开始下降

4 8～72h接近正常水平。结论:脑缺血再灌注损伤可以诱导VEGF表达增强

提示VEGF对缺血性脑损伤有保护作用

Abstract

Objective:To study the expression of vascular endothelial growth factor (VEGF) and VEGF mRNA after focal cerebral ischemia reperfusion in rats.Methods:MCAO (Middle Cerebral Artery Occlusion) models were set up in rats

and the rats were divided into six groups:control group

5 groups of ischemia for 2h then respective reperfusion for 6h

12h

24h

48h and 72h.The samples were analyzed with immunohistochemical methods.The expression of VEGF mRNA was detected by RT-PCR analysis.Results:VEGF protein expression started at 6h.It increased remarkably at 24h

and it was strongest at 48h.VEGF mRNA increased at 6h

while reached peak value at 12h and reduced at 24h

finally returned to normal level at 48-72h.VEGF expression was mainly detected in the periphery of infarcts at every timepoint.Conclusion:These findings demonstrate that the VEGF strong expression can be induced in focal cerebral ischemia reperfusion in vivo.The changes of VEGF levels suggest that VEGF has neuroprotective effect against cerebral ischemic injury.
