摘要

目的 :研究人参总皂甙（TSPG）诱导K5 6 2细胞凋亡的机理。方法 :采用形态学观察、原位末端标记法（TUNEL）观察不同浓度人参总皂甙对K5 6 2白血病细胞凋亡的影响 ;用免疫组化法检测TSPG诱导K5 6 2细胞凋亡中Fas表达情况

并采用酶联免疫吸附法（ELISA）检测可溶性Fas（sFas）。结果 :5 0、10 0 μg/mlTSPG可诱导K5 6 2细胞凋亡 ;同时实验组Fas表达明显增高（P <0 .0 1）

而sfas无明显变化。结论 :TSPG诱导K5 6 2细胞凋亡与其诱导细胞Fas高表达有关。

Abstract

Objective:Our purpose is to clarify the mechanisms for total saponins of Panax ginseng(TSPG) inducing K562 cells to apoptosis.Methods:By using morphological observation and TUNEL

the effect of TSPG on apoptosis of K562 cells were studied.The expression of Fas was studied by immunocytochemistry

and sfas was assayed by ELISA.Results:The results indicated that 50

100μg/ml TSPG could induce K562 cells to apoptosis.Our experiment also showed after induced by TSPG

the ratio of positive K562 cells of expression Fas is increased ( P <0.01)

while sfas had no change.Conclusion:The mechanisms for TSPG inducing K562 cells to apoptosis may be related to the increased expression of Fas in K562 cells.
