Objective :To study the effects of ascorbic acid(vitamin C) on PKC -β1 activity in kidneys of diabetic rats. Methods :30 Wistar rats were randomly divided into three groups: Control group (group C)
streptozotocin - induced diabetic rats group(group 13) and diabetic rats treated with ascorbic acid 90mg/(kg·d)(group DT). The concentration of ascorbic acid in plasma and kidney were measured at 9 week. The activity of PKC - β1 was observed by immunofluorescence technique and laser confocal microscope. Results : Compared with group C
the level of ascorbic acid in the plasma and kidney of group D decreased significantly (P<0.05)
and the activity of PKC - β1 was markedly increased. After 8 weeks of treatment with ascorbic acid
the PKC - β1 activity was nearly normalized and the level of ascorbic acid in the plasma and kidney was increased significantly. Conclusion : This finding suggests that the increased PKC - ft activity in kidneys of diabetic rats may be associated with lower levels of ascorbic acid. Administration of ascorbic acid can inhibit the PKC- β1 activation in kidneys of diabetic rats.
