Objective: Exploring the expression and significance of cell apoptosis and apoptosis genes of varico-sity in the lower extremities. Methods:By using TUNEL
hybridization in situ
immunohistochemical method
83 patients with varicosity and 10 controls were studied. Results: Apoptosis index of ECs and SMC in cystic dilata-tions were compared with those of non-cystic dilatations and controls with significant difference (P <0.01). The numbers and index of Bcl-2 mRNA positive ECs and SMC expression were greatly lower in cystic dilatations than those in non-cystic dilatations (P<0.05). Bcl-2 mRNA expression in non-cystic dilatations were markedly higher than that in controls (P<0.05). The numbers and index of protein Bcl-2 positive Ecs and SMC were significantly increased in non-cystic dilatations compared with those of cystic dilatations and control (P <0.05). The numbers and index of protein Bax and Fas positive expression increased markedly in cystic dilatations compared with those of non-cystic dilatations and control (P<0.05). Expressions of Bcl-2 mRNA were positively correlated with that of protein Bcl-2 (r=0.795381
P=0.001981). Conclusion: Over-apoptosis led to venous remode-ling may be one of the mechanism of varicosity. Over-apoptosis of ECs and SMC in cystic dilatations may correlate with low level expression of Bcl-2 mRNA and protein
