Objective: To observe the role of taurine on apoptosis of isolated pancreatic islet cells induced by combination of IL-1β
TNF and IFN-γ.Methods: Isolated pancreatic islet cells from Wistar rat were incubated in monolayer in vitro.NO production
NOS activity
apoptotic percentage of islet cells and DNA fragmentation in pancreatic islet cells incubated with combination of IL-1β
TNF and IFN-γ, taurine were measured .Results:Combination of IL-1β
TNF and IFN-γ could induce a significant increase in apoptotic percentage of pancreatic islet cells and DNA fragmentation.NO production and NOS activity also markedly increased (P<0.01),which were blocked by addition of taurine to the pancreatic islet cells (P<0.01)。Conclusion:NO might be one of the important ways of inducing apoptotic cell death of pancreatic islet cells by a combination of IL-1β
TNF and IFN-γ.Taurine attenuates the cytokines-induced β cells apoptosis, the mechanism of which may be the inhibition of NOS and the decreasing of NO production.
