P53 overexpression and mutation in endometrial carcinoma[J]. Journal of Chongqing Medical University, 2000,(4):362-364. DOI: 10.13406/j.cnki.cyxb.2000.04.011.
:Objective To investigate p53 gene mutations in endometrial carcinoma(EC)
and their correlation with clinical features and prognosis. Methods Using immunohistochemical staining(IMH)and polymerase chain reaction-single strand conformation polymorphism(PCR-SSCP)methods
examine the protein expression and gene mutation of p53 in EC. Results The general positive rate of P53 protein was 34.15%. The expression of p53 was not correlated with age, clinical stage, myometrial invasion ,lymph node or ovarian metastasis(P>0.05),but the pathologic grade(P<0.05).Survival curves determined by Kaplan-Meier method showed that p53 overexpression was associated with a poor prognosis in EC(P<0.05).The positive rates of p53 gene mutation was 23.08% in EC
and the mutation were located on exon 5,6,and 7.The concordance rate of IMH and PCR-SSCP was 76.92%. Conclusion The mutation of p53 gene were associated with the pathologic grade and poor prognosis in EC.
