摘要

用葡聚糖凝胶Sephadex G-200柱层析法、三氯醋酸-乙醇提取物DEAE-Cellulose（DE52）柱层析法、尿素提取物Sepharose 4B柱层析法对华支睾吸虫成虫可溶性抗原进行部分纯化

获得8种不同的抗原活性组分.并用免疫电泳、聚丙烯酰胺凝胶圆盘电泳（PAGE）、薄层等电聚焦电泳、微板法动力学酶联免疫吸附试验进行鉴定.认为经纯化的抗原组分较粗抗原更适宜于临床诊断及考核疗效

Abstract

Crude adult worm antigen (AWA) of Clonorchis sinensis was purified by three different procedures respectively: 1.AWA pass through sephadex G-200

2. trichloroacetic acid soluble alcohol insoluble composition pass through DEAE- cellulose (DE-52)

3. 8M urea soluble composition pass through sepharose 4B. 8 different purified antigen fractions were obtained. Their purity

antigen activity and responsive antibody activity were identified by immunoelectrophoresis

polyacrylamide gel disc electrophoresis

isoelectric focusing

kinetic -dependent enzyme-linked immunosorbent assay (K-ELISA) respectively. The results indicated that the purified antigen fractions were more effective than the crude antigens for diagnosis and evaluating therapeutic efficiency in clinical practice
