The GABA hypothesis for the pathogenesis of hepatic encephalopathy suggested bySchafer in 1982 had been supported by quite a lot of experimental data. However
up to now it is still unknown whether the brain uptake of blood GABA is increased or not in liver failure. For this purpose
we performed tracing experiments in mice with galactosami-ne-induced hepatic encephalopathy. After injection of 3H-GABA
the radiolabelled GABA in mice brain and serum were seperated from its metabolites by cellulose acetate electropho-resis and the redioactivities were determined(Serum/Total%; Brain/Total%). The ratio of 3H-GABA to total non-volatile radioactive compounds in serum and brain were also determined (Serum 3H-GABA%; Brain 3H-GABA%). Our results clearly indicated that Serum/ Total%
Brain/Total%
Serum 3H-GABAfo
and Brain 3H-GABA% of mice wisth gala-ctosamine-induced hepatic encephalopathy were significantly higher than that of the normal. It was concluded that the brain uptake of GABA from blood was increased in mice with experimental hepatic encephalopathy and it may be the result of the change of permeability of blood brain barrier to GABA in liver failure
