Objective:To explore the effect and mechanism of Wnt/β-catenin signaling pathway in H9 c2’s cell adaptation to chronic hypoxia. Methods:Embryonic rat-heart-derived H9 c2 cells were exposed to 37 ℃
5%O2
5%CO2
and 90%N2 as the experimental?group;cells were collected at 6
12
24
36
48
72 h. The cells were cultured under 37 ℃
21%O2
5%CO2 as control group. Expression of β-catenin
bcl-2 and survivin m RNA or protein at different time points were measured by using Western blot and real-time PCR.Results:Protein levels of β-catenin increased at 6
12
24
36 h compared with those of control group(P=0.000
P=0.018
P=0.001
P=0.000). bcl-2 and survivin were significantly increased after β-catenin reaching high level of expression(P =0.000
P =0.000).Conclusion:Activation of Wnt/β-catenin signaling pathway may contribute to H9 c2 cell adaptation’s to chronic hypoxia
partly through upregulating anti-apoptosis genes expression to promote cell survival.
