Objective:To observe the effects of curcumin on the growth and vascular growth factors in Daoy cells
and to explore the potential mechanisms. Methods:Human medulloblastoma Daoy cells were cultured in vitro
and treated with different concentrations of curcumin(5
10
20 and 40 μmol/L) for different times(24
48
72 h). MTT assay was used for detecting the effects of curcumin on the proliferation of Daoy cells. ELISA was applied to detect the changes of VEGF levels in the supernatant
and Western blot was used for determining the expression of β-catenin of Wnt/β-catenin pathway and vascular endothelial growth factor(VEGF) at protein levels. Results :MTT results showed that curcumin significantly inhibited the proliferation of Daoy cells
and they were in a concentration-and time-dependent manner. ELISA results demonstrated that curcumin could decrease the generation of VEGF and Western blot indicated that curcumin not only reduced the expression of VEGF
but also reduced the expression of β-catenin
they were also in a concentration-and timedependent manner. Conclusion:Curcumin could inhibit the proliferation of mdulloblastoma Daoy cell
the mechanisms may be related to the inhibitory action of curcumin on Wnt/β-catenin pathway and its downstream VEGF
which will provide a novel theoretical basis for curcumin treating with medulloblastoma.
