摘要

目的:检测Nme2（nucleoside diphosphate kinase 2）基因在小鼠早期妊娠和人工诱导蜕膜化的子宫内膜组织中的表达规律;探究Nme2基因与子宫基质细胞蜕膜化的相关性。方法:利用免疫组织化学、Western blot和实时荧光定量PCR（q RTPCR）分别检测小鼠妊娠第5天着床点（D5IS）、着床旁（D5IIS）

D6IS、D6IIS、D8IS、D8IIS及人工诱导蜕膜化模型小鼠子宫内膜中Nme2的表达定位

m RNA和蛋白质水平的表达水平。结果:小鼠D5、D6、D8子宫内膜中着床点Nme2基因的m RNA和蛋白水平的表达明显高于着床旁;免疫组织化学结果显示Nme2表达定位表达于D5着床点的初级蜕膜区（primary decidua region

PDZ）、D6和D8着床点的次级蜕膜区（secondary decidua region

SDZ）;人工诱导蜕膜化模型中

Nme2基因的m RNA和蛋白质水平在诱导组的表达明显高于对照组。结论:Nme2基因的表达上调与小鼠基质细胞的蜕膜化过程相关。

Abstract

Objective:To investigate the expression profile of Nme2 in the endometrium of early pregnant mouse and artificially induced pregnant mouse and to explore whether Nme2 is involved in the decidualization of stromal cells. Methods:Immunohistochemistry

Western blot and real-time quantitative PCR(q RT-PCR)were used to detect the expression of Nme2 in mouse endometrium from implantation sites of pregnant day 5(D5IS)

inter-implantation of pregnant day 5(D5IIS)

D6 IS

D6IIS

D8 IS

D8IIS and artificially induced deciduaoma. Results:The expression of Nme2 m RNA and protein in the D5

D6 and D8implantation sites was significantly higher than that of the inter-implantation sites. Immunohistochemical results showed that Nme2 staining was localized in the primary or secondary decidualization zone of D5

D6 and D8implantation sites. Moreover

the expression of Nme2 m RNA and protein in artificially induced deciduaoma was also significantly higher than that in the control. Conclusion:Nme2 expression is associated with decidualization in mouse endometrium.
