摘要

目的:研究Wnt10b对体外培养小鼠触须毛囊生长的作用。方法:构建Dct-Lac Z小鼠触须毛囊体外培养模型。用腺病毒Ad-Wnt10b和Ad-GFP感染触须毛囊2 d、4 d并对触须生长长度进行测量。X-gal染色检测触须毛囊中黑素细胞数量。Western blot检测酪氨酸相关蛋白1(tyrosinase-related protein 1

TRP-1)、酪氨酸酶(tyrosinase

TYR)蛋白的表达水平。结果:腺病毒感染4 d后长度测量结果显示Wnt10b处理组的触须毛囊生长长度明显高于GFP对照组(0.58±0.03 vs.0.09±0.05

P=0.000);X-gal染色结果显示Wnt10b处理组的黑素细胞数量明显多于GFP对照组;Western blot结果显示Wnt10b处理组的触须毛囊中TRP-1和TYR的蛋白表达明显升高(1.14±0.16 vs.0.55±0.22

1.34±0.33 vs.0.77±0.09;P=0.020

P=0.045)。结论:Wnt10b能够促进小鼠触须毛囊的体外生长。

Abstract

Objective:To investigate the effect of Wnt10 b on the growth of mouse whisker follicles in vitro.Methods:Dct-Lac Z mouse was used to establish the model in vitro for the growth of whisker follicles.After Whisker follicles being infected with Ad-Wnt10 b and Ad-GFP for 2 days and 4 days

the length of whisker was measured

the melanocytes was observed by X-gal staining and the protein expression of tyrosinase-related protein 1（TRP-1）and tyrosinase（TYR）were measured by Western blot.Results:After being infected with adenoviruses for 4 days

the average growth length of Wnt10 b group was longer than that of GFP control group（0.58±0.03 vs.0.09±0.05

P<0.001）;the number of melanocytes of the Wnt10 b group was more than that of the GFP control group.Wnt10 b significantly increased the protein expression of TRP-1 and TYR in the whisker follicles compared with that of GFP control group（1.14±0.16 vs.0.55±0.22

P=0.045）.Conclusion:Wnt10b may promote the growth of mouse whisker follicles in vitro.
