Objective:To detect the expression of non-muscle myosin heavy chain 9(MYH9)in human hepatocellular carcinoma(HCC)and its role in cell invasion. Methods:q RT-PCR and immunohistochemistry was applied to detect the expression of MYH9 in HCC(n=65)and matched tumor-adjacent tissues. The correlation between MYH9 expression and clinical features was analyzed by Pearson chi-square test. si RNA was used to knock-down the expression of MYH9 in MHCC-97 H cells. The effects of MYH9 on cell migration and invasion were measured by wound healing assay and transwell assay
respectively. Results:Both the m RNA and protein expression of MYH9 were significantly up-regulated in HCC tissues
and the positive expression of MYH9 was associated with the number of nodules(≥2)and advanced TNM stage(Ⅲ+Ⅳ stage). Down-regulating the expression of MYH9 could inhibit migration and invasion in MHCC-97 H cells. Conclusion:The expression of MYH9 is significantly higher in HCC tissues. Down-regulation of MYH9 could achieve an anti-cancer role through inhibiting cell migration and invasion.
