Objective:To investigate the effect of Curcumin on the migration of human umbilical vein endothelial cells(HUVECs)induced by oxidized low-density lipoprotein(ox-LDL). Methods:The HUVECs were cultured in vitro and treated with ox-LDL in different concentrations for 24 h;the apoptosis rate was determined by flow cytometry. Cells were divided into three groups: normal control group;ox-LDL treatment group;curcumin pretreatment+ox-LDL group. The concentration of reactive oxygen species(ROS) was measured by laser confocal microscopy;the level of nitric oxide(NO)was dectected by method of colorimetry;the protein levels of endothelial nitric oxide synthase(e NOS)
phosphorylated e NOSS1177(p-e NOSS1177)
protein kinase B(Akt) and phosphorylated Akt S473(p-AktS473)were detected by Western blot;the migration ability of endothelial cells was measured by transwell method. Results:High concentrations(≥40 mg/L)of ox-LDL increased the apoptosis rate significantly(P=0.010). Compared with those of control group
the level of ROS(P=0.001)was obviously increased by ox-LDL
phosphorylation of Akt S473
e NOSS1177
concentration of NO and the migration ability significantly were inhibited(P=0.000
P=0.000
P=0.000). Compared with those of ox-LDL treatment group
the generation of ROS(P<0.05)was reduced by curcumin pretreatment
and the phosphorylation of Akt S473
e NOSS1177
the concentration of NO(P<0.05
P<0.05
P<0.05)and the ability of migration were increased significantly(P<0.05). Conclusion:Curcumin inhibits the production of ROS induced by ox-LDL
and may increase the phosphorylation of Akt S473 and e NOSS1177 to promote the ability of endothelial cell migration.
