摘要

本文应用ＰＣＲ技术从ＨＢｅＡｇ单克隆抗体鼠─鼠杂交瘤细胞基因组ＤＮＡ中扩增出重链可变区基因（ＶＨ），并将ＶＨ基因与ｐＵＣ１９载体连接，转化ＪＭ１０９菌，克隆出ＶＨ基因。重组质粒ｐＵＣ－ＨＢｅＡｇＶＨ经荧光核酸测序表明，该ＶＨ基因大小为３２７ｂｐ，编码１０９个氨基酸。

Abstract

The VH gene ws amplified from genomic DNA of hybridomacells cells producing McAb against HBeAg byPCR reaction. The PCR products were ligated with PUC19 vector.Then VH gene was cloned by transforming itinto JM109. The recombinant plasmid pUC─HBeAgVH was sequenced by chain terination─based cyclesequencing protocal. The result shows that the VH gene was 327 base pair encoding 109 ainino acids.
