Objective: To culture microvascular endothelial cells from Wistar rat cerebral cortex. Methods: Cerebral cortex from Wistar rats of 1~5days filtered by various diameter meshes. The pellets containing the microvessels were washed and collected by centrifugation.The microvessels obtained from above procedures were Ⅶdigested by collagenase .The isolated endothelial cells were cultured in M199 +15% fetal calf serum at a density ×of 3104/ml and observed morphologically. Results:The cultured microvascular endothelial cells were stained Ⅷpositively for factor relative antigen. On the 7~9th day

the cells displayed monolayer growth and looked like "cobblestones"

a typical appearance of cultured endothelial cells under inverted microscope. Conclusion: It is a simple and easy method to culture endothelial cells from brain microvessels.
