Objective:To observe the prevention and therapeutic effects of simvastatin on the osteoporosis in rats.Methods:Forty SD female rats were randomly divided into four groups:sham operation group

OVX group

estrogen group and simvastatin group.One month after OVX operation

we began medicine intervention.Estrogen group was subcutaneously injected EB(0.1mg/kg)per triduum until being sacrificed.Simvastatin group was intragastrically administered simvastatin 5mg/(kg.d)for 2 weeks

then the administration was stopped for 5 weeks and then continued for 2 weeks until being sacrificed.After the intervention

we observed the change of rats’ BMD

bone histopathology and expressions of IL-6 and BGP.Results:The level of BMD in OVX group was lower than that in the sham group(P<0.05).The level of BMD raised in treated groups compared to the OVX group(P<0.05).The expression of IL-6 in OVX group raised compared to the sham group(P<0.05).The expression of IL-6 in all treated groups degraded compared to the OVX group(P<0.05)

especially in the estrogen group.The expression of BGP in OVX group raised compared to the sham group(P<0.05).The expression of BGP in all treated groups

especially the simvastatin group

significantly raised compared to the OVX group(P<0.05).The change of bone histopathology in treated groups was ameliorated compared to the OVX group.Conclusion:Simvastatin could effectively prevent and cure osteoporosis by elevating BMD

ameliorating the bone histopathology

raising the expression of BGP and degrading the expression of IL-6.
