Objective: To investigate the clinical significance of the expressions of survivin gene and its main transcript variants mRNA in colorectal carcinoma. Methods: The expression of total survivin
survivin-△Ex3
survivin-2B
survivin-3B
survivin 2α
and CEA CAM 5 mRNA in 39 cases colorecta1 cancer
adjacent tissues (2.5 cm from the cancer margin) and norma1 colorecta1 tissue (10 cm from the cancer margin) were detected by RT-PCR technique and semi-quantitative methods use gel imaging analytical system or SYBR GreenⅠreal-time fluorescent PCR. Results: The positive rates of total survivin
survivin-△Ex3
survivin-2B
survivin-3B
survivin2α
and CEA CAM5 mRNA in colorectal carcinoma and adjacent tissue were 100.0%

showing significant difference in total survivin
survivin-△Ex3 and survivin-2B from adjacent tissues (P<0.05). Total survivin
four transcript variants and CEA CAM5 mRNA levels were higher in colorectal carcinoma than in adjacent tissue (P<0.01);but no significant difference was found between adjacent carcinoma and norma1 colorecta1 tissues (P>0.05). Several transcript variants mRNA levels were associated with some clinicopathologic parameters (P<0.05). Conclusion: The survivin and its transcript variants mRNA were highly expressed in colorectal carcinomas
several transcript variants mRNA levels were associated with clinicopathologic parameters. Detection of Survivin transcript variants mRNA expression may be more valuable than the CEA mRNA in early prediction of colorectal carcinoma.
