Objective:To investigate the efficacy and safety of S1
oxaliplatin
irinotecan and leucovorin combination chemotherapy in advanced gastric carcinoma.Methods:48 patients with advanced gastric cancer were enrolled.Patients receiving S1
at 80 mg/m2
was orally administered twice daily for 2 weeks
followed by a drug-free interval of 1 week(one cycle).Concurrently
Irinotecan(150 mg/m2) and leucovorin(200 mg/m2) were given on day 1
twice daily for two weeks
followed by oxaliplatin 130 mg/m2.The chemotherapy was repeated every 4 weeks. All patients received 2 cycles at least.Efficacy and toxicities were evaluated according to the RECIST 1.1 standard. Results:All the 48 patients were assessable for response. There were 5(10.4%) complete remission(CR)
23(47.9%) partial remission(PR)
12(25.0%) stable disease(SD)
and 8(16.7%) progression disease(PD).The overall response rate was 58.3%.The disease control rate(DCR) was 83.3%.Median time to progression(TTP) and median survival time(MST) were 8.9 and 12.3 months
respectively. The major toxicities were neurosensory abnormity
leukopenia
nausea
hematologic toxicity and vomiting. Conclusion:The combination of S1
oxaliplatin
irinotecan and leucovorin has good clinical activity with acceptable toxicity in patients with advanced gastric cancer.
