摘要

目的:初步探讨肿瘤坏死因子α转换酶（Tumor necrosis factor-αconverting enzyme

TACE）在小鼠动情周期中的表达规律。方法:用RT-PCR及免疫组化方法分别检测动情周期各期小鼠子宫内膜TACEmRNA和蛋白的表达。结果:TACEmRNA在动情周期小鼠子宫内膜呈规律性表达

在动情期表达明显增强

与其它3期相比有显著性差异（P﹤0.01）;其它3期间无显著性差异（P>0.05）。免疫组化分析显示TACE在子宫内膜的表达规律与RT-PCR一致。结论:TACE通过裂解EGFR家族多种配体释放其胞外区

从而激活EGFR信号途径

在子宫内膜的周期性变化中起着重要作用。

Abstract

Objective:To primarily investigate the expression pattern of tumor necrosis factor-α converting enzyme(TACE) in mouse endometrium during the estrus cycle.Methods:Reverse transcription polymerase chain reaction(RT-PCR) and immunohistochemistry were used to detect the expression of TACE mRNA and protein in mouse endometrium during each stage of the estrus cycle.Results:The expression of TACE mRNA in mouse endometrium during estrus cycle had regular pattern.The TACE mRNA/β-actin mRNA expression in estrus stage was higher than those of the other 3 groups

and there was significant difference between estrus stage and the other 3 groups(P﹤0.01).Besides

there was no significant difference among the other 3 groups(P>0.05).Immunohistochemical analysis showed the same expression pattern of TACE in endometrium as RT-PCR.Conclusion:TACE may play a key role in cyclic changes of normal endometrium through proteolytic removal of the extracellular domain of multiple ligands of EGFR family to release their extracellular domains which is capable of activating EGFR signal way.
