摘要

目的:比较两种多囊卵巢综合征（Polycystic ovary syndrome

PCOS）大鼠模型的构建方法。方法:模型组&10只大鼠使用脱氢表雄酮（DHEA）+胰岛素（INS）造模;模型组’10只大鼠使用绒毛膜促性腺激素（HCG）+胰岛素（INS）造模

主要观察指标为阴道脱落上皮细胞涂片

卵巢组织学改变

血清性激素水平

空腹胰岛素值及空腹血糖值并计算HOMA指数（HOMA-IR）。结果:组)、组*中失去规律动情周期的大鼠的比例分别为90%、100%;两组大鼠卵巢重量显著高于相应对照组;两组大鼠卵巢均可见多个扩张的囊泡

平均囊泡直径组-（0.048±0.012）显著小于组/（0.069±0.036）

黄体计数组1（0.10±0.32）显著少于组3（0.70±0.82）;两组与相应对照组大鼠相比较血清T水平、LH/FSH比值均有显著性升高

E2升高不明显（P>0.05）;两模型组胰岛素抵抗倾向显著（P<0.05）

HOMA-IR模型组5（8.89±0.86）显著高于模型组7（6.86±0.48）。结论:脱氢表雄酮+胰岛素的造模方法在卵巢形态学改变及胰岛素抵抗程度等指标上较符合目前PCOS的特征。

Abstract

Objective:Tow kinds of Polycystic Ovary Syndrome models in rats were compared.Methods:In the model ⅰ

ten female rats were given DHEA+INS.In the model ⅱ

ten female rats were given HCG+INS.Then the ovulation

morphology of ovary

blood serum levels of sexual hormones

fasting plasma glucose(FPG)

fasting insulin(FINS) in the two models were assyed

Results:There were 90 percent of rats losting regular estrous cycle in the model ⅰand 100 percent of rats in the model ⅱ.The ovary weight of rats in both two models was heavier than that of controls.Rats of both two models showed polycystic ovary-like syndrome manifestation

The average saccular ovulas in model ⅰ(0.048±0.012) was smaller than those in model ⅱ(0.069±0.036)

The number of corpora lutea in modelⅰ(0.10±0.32) was smaller than that in model ⅱ(0.70±0.82).Serum levels of T

LH/FSH increased in both two model.Serum levels of E2 had no significant increase(P>0.05).HOMA-IR of both two models were higher than that of controls with model ⅰ(8.89±0.86) significantly higher than model ⅱ(6.86±0.48). Conclusion:The model of DHEA+INS is more similar to human PCOS in the morphology of ovary and the degree of insulin resistance.
