Objective:To investigate effects of dendritic cells(DCs) pulsed with autologous hepatoma cell lysates on the immune function of the postoperative patients with hepatocellular carcinoma(HCC).Methods:DCs isolated from peripheral blood mononuclear cell of HCC patients were cultured and proliferated in vitro by using rhGM-CSF and rhIL-4
and then were pulsed with autologous hepatoma cell lysates.30 postoperative patients with HCC were randomly divided into 2 groups.15 cases were treated with dendritic cell vaccine.15 cases received only chemotherapy and were established as control group.Immune function、clinical effects in two groups of patients were compared and observed. Results: The levels of DC3~+、DC4~+/CD8~+ and NK cell in the DC vaccine group significantly increased after vaccination
while those of the control group had no significant changes. The levels of IgG、IgM、IgA in the DC vaccine group increased after vaccination
but didn’t reach statistical significance. The concentration of IL-10 in the DC vaccine group significantly decreased after vaccination(P<0.05).Conclusion:The hopatome cell lysates pulsed DC vaccines can improve the immune function of the postoperative patients with HCC.This method results in fewer side effects and should be recommended.
