The pharmacodynamics of diazepam solution administered rectally for seizure control in rats and children [J]. Journal of Chongqing Medical University, 1998,(1).DOI:
In order to investigate the practical value of rectal administration of diazepam solution
we studied the blood levels and pharmacodynamics of a single dose of diazepam solution administered rectally to Wistar rata and to children for seizure control. The results showed:(1)Diazepam was absorbed rapidly from the rectum in rats and children. 5 minutes after administration
the blood level of diazepam was over the diazepam therapeutic level necessary for seizure control
tmax was 12.5± 2.5 and 18.1±10.0 minutes.(2)5 minutes later
significant activity was present in the EEG recordings of both the rata and children. indicating that diazepam had an effect on their cortex. The frequency of epileptiform discharges in the EEG of the epileptic patients was obviously reduced. Compared with the control group
the experimental seizures were effectively prevented.(3)There were no visble pathological alterations on rectum. No side effects were found in the rata or children. Conclusion; Rectal administration of Diazcpqm is a safe and effective method for seizure control. It can be used easily and conveniently in cases out of hospital to prevent hypoxemic brain damage due to prolonged seizure.
