摘要

目的：观察不同时期缺血心肌中VEGF mRNA及蛋白质表达及毛细血管新生的变化规律。方法：雄性Wistar大鼠30只，分别检测心肌梗塞后24h、1W、2W、3W、4W缺血心肌中内皮细胞数，毛细血管密度和VEGF mRNA及蛋白质表达情况。结果：（1）心肌梗塞1周后缺血心肌中毛细血管密度、内皮细胞数均明显增加，2周后达高峰，3周后开始下降， 4周后下降到1周时水平。（2）心肌梗塞24h后，VEGF mRNA表达开始增加，1周后达高峰，2周后开始逐渐下降，4周后几乎无表达；VEGF蛋白质的表达从1周后增加，2周后达高峰，3周后开始下降，4周降低到1周时水平。结论：心肌梗塞后缺血心肌中毛细血管密度、VEGF mRNA及蛋白质表达均显著增加；缺血心肌中毛细血管密度与VEGF表达的变化相一致。

Abstract

Objective: To investigate the changes of expression of VEGF and angiogenesis in ischemic myocardium of rats in various periods. Methods: 30 male infarcted rats were divided into 24h group

1w group

2w group

3w group and 4w group. The myocardial samples from all groups were taken for detecting the numbers of endothelial cell (EC)

capillary density (CD)

and expression of VEGFmRNA and protein. Results : ⑴ECs and CD increased at 1 week after AMI

and peaked at 2 weeks after AMI

while gradually decreased at 3 weeks after AMI

decreased at 4weeks after AMI to the same level with that at 1 week after AMI.⑵Expression of VEGFmRNA in ischemic myocardium increased at 24 hours after AMI

peaked at 1 week after AMI

gradually decreased at 2 weeks after AMI

and little expression was found at 4 weeks after AMI. Expression of VEGF protein increased at 1 week after AMI

peaked at 2 weeks after AMI

gradually decreased at 3 weeks after AMI

decreased at 4 weeks to the same level with that at 1 weeks after AMI. Conclusion : CD

expression of VEGF increased in ischemic myocardium of infarcted rats

and the changing trend of these two variables is comparable.
