Objective:To explore the mechanisms of angiogenesis
invasion and metastasis inhibited by arsenic trioxide(As2O3)in human renal cell carcinoma 786-0 cells. Methods:The changes of VEGF
nm23
MMP-9
and TIMP-1 protein expression were studied by immunocytochemistry
and TGF-β1
TβR-I and TβR-II mRNA were analysed by reverse transcription polymerase chain reaction in 786-0 cells treated with As2O3.Results:2μmol/L As2O3 effectively inhibited the protein expression of VEGF and MMP-9 in 786-0 cells(P<0.05)
but increased nm23 and TIMP-1 protein expression(P<0.05).Absence of expression of TβR-ImRNA was found in 786-0 cells
and no change of mRNA expression of TGF-β1 and TβR-II was detected in 786-0 cells treated with As2O3 for 72h.Conclusions As2O3 might inhibit the angiogenesis
invasion and metastasis of 786-0 cells by down-regulating the expression of VEGF and MMP-9 protein and up-regulating nm23 and TIMP-1 protein.
