Objective:To investigate the pathogenesis of active oxygen scavenger(catalase
CAT) and methylprednisolone sodium succinate(MP) on experimental demyelinating optic neuritis. Methods:The expenimental rats received intraperitoneal injections of CAT
physiologic saline(PS) and MP after antigenic sensitization. On day 7th and 14th after antigenic sensitization
H2O2 measurement
patho-image analysis
and histiocyte ultrastructure analysis were applied to study the effects of CAT and MP on experimental demyelinating optic neuritis. Results:The concentration of H2O2 in optic nerves of experimental demyelinating optic neuritis was higher than that in the normal optic nerves.CAT and MP can reduce not only the content of H2O2 in optic nerves of EAE rats
but also the number of inflammatory cells and reactive astroglial cells
the extent of demyelination in the retrobulbar optic nerves. But the effect of MP on H2O2 scavenging and myelin protection was weaker than that of CAT.Conclusion:H2O2 may has some relationship with the occurrence of experimental demyelinating optic neuritis.
