Objective:To investigate the expression of α-caten in and focal adhesion kinase(FAK)in colon carcinoma and to reveal their correlation with tumor clinicopathological features. Methods:The expression of α-catenin and FAK in tumor tissue and normal tissue of 34 colon carcinoma patients were detected by immunohistochemistry and Western blot. Results:The expression of α-catenin and FAK were detected both in tumor tissue and normal tissue
mostly in plasmalemma and endochylema. The expression of α-catenin was significantly lower in tumor tissue than that in normal tissue(t=10.693
P<0.01)
but the expression of FAK was higher in tumor tissue than that in normal tissue(χ2=10.350
P=0.001). A weak negative correlation was found between FAK and α-catenin(Pearson correlation coefficient=-0.383
P=0.025).The expression of α-catenin and FAK were significantly correlated with TNM staging、distant metastasis and lymph node metastasis of colon carcinoma (P<0.01)
but not with the sex and age of patients. The expression of α-catenin was also related to tumor differentiation status
but the expression of FAK was not statisticaly significant in different tumor differentiation status. Conclusion:The overexpression of FAK and the reduced expression of α-catenin in tumor tissue play an important role in the tumorigenesis and progression of colon carcinoma.
