Objective:To prepare pshRNA-survivin liposome with high entrapment efficiency and good stability

and evaluate its quality.Methods:pshRNA-survivin liposome was prepared by the reversed-phase evaporation method.The optimum formation was selected by means of orthogonal design of experiment;particle size

zeta potential and stability of the liposome were evaluated.Entrapment efficiency was determined by centrifugal ultrafiltration.Centrifugal acceleration experiment and determination of leak rate were served to prove the liposome stability.Results:The optimum formula was as follows:the ratio of lecithin to pshRNA-survivin was 100∶1;oil-water ratio was 4∶1;lecithin:cholesterol was 1.5∶1;temperature of steaming was 37℃;the average diameter and zeta potential of pshRNA-survivin liposome were 123.4 nm and(-8.42±0.88) mV.The average entrapment efficiency for pshRNA-survivin was 77.9%.Conclusion:The selected formulation and preparation technique of pshRNA-survivin liposome is reasonable in prescription
