Development and validation of simultaneous recording of cystometrography and external urethral sphincter electromyography in mice
To establish and validate simultaneous recording of cystometrography (CMG) and external urethral sphincter electromyography (EUS EMG) in mice.
Methods
2
Twelve female
age-matched C57BL/6 mice were anesthetized
and the pudendal canal was located bilaterally
followed by implantation of a polyethylene catheter into the bladder for simultaneous recording of CMG
leak point pressure (LPP) and EUS EMG. After bilateral pudendal nerve transection (PNT)
the LPP and EUS EMG recording was repeated on the same mice. Statistical comparisons of quantitative data before and after PNT were made using a paired
t
-test with
P
<0.05.
Results
2
When the pudendal nerve was intact
EUS EMG amplitude and frequency were increased significantly in response to LPP testing (from 38.9±8.4 μV to 111.7±19.4 μV and from 37.6±9.5 Hz to 142.6±15.0 Hz
respectively
P
<0.001). Quantitative analysis of EUS EMG bursting activity during voiding was characterized with intrabursting amplitude of 70.4±15.2 μV
intrabursting firing rate of 393.8±118.9 Hz
interbursting interval of 222.0± 44.2 ms and bursting duration of 85.60±6.21 ms. Bilateral PNT eliminated EMG activity
which verified the recording quality. Besides
LPP
peak bladder pressure and maximum bladder pressure during voiding were also significantly reduced after PNT (16.4±1.2
vs
7.4±0.7 cmH
2
O
P
<0.001; 26.7±1.7
vs
15.9±1.1 cmH

Conclusion
2
Simultaneous recording of CMG and EUS EMG in mice is feasible
so it can be used as functional outcome in the investigation of lower urinary tract symptoms.
