Inhibiting TIM-4 function in Kupffer cells induces iTreg cell proliferation and thus immune tolerance in mice after liver transplantation
To determine the effect of inhibiting T-cell immunoglobulin and mucin-domain-containing molecule 4 (TIM-4) function in Kupffer cells (KCs) on the proliferation in iTreg cells after liver transplantation in mice and the relevant mechanism.
Methods
2
The mouse models of orthotopic liver transplantation were established
and the mice were divided randomly into 3 groups: sham group
control mAb group
and TIM-4 mAb group. The number of activated KCs was assayed by flow cytometry. The expression of TIM-4 in the liver tissues was assayed by Western blotting
and the apoptotic index (AI) was analyzed by TUNEL assay. KCs were extracted from the liver. The expression of TIM-4 in the KCs was observed by laser confocal microscopy. The KCs and naive CD4
+
T cells were co-cultured
and then divided into 3 groups: control group
control mAb group
and TIM-4 mAb group. The proliferation of naive CD4
+
T cells was assayed with CellTrace CFSE cell proliferation kit. Furthermore
the number of CD4
+
CD25
+
Foxp3
+
T cells was assayed by flow cytometry. The levels of IL-4
IL-6 and IL-13 were assayed by ELISA
and the protein level of p-STAT6 in T cells was assayed by Western blotting. Then
the mice in the iTreg group were injected with iTreg cells
which were induced with co-culture system of KCs treated with TIM-4mAb and naive CD4
+
T cells through portal vein. The sham group and liver transplantation (LT) group were treated with commensurable PBS. In 7 d after operation
the serum levels of AST
ALT and TBIL were tested in each group. The liver tissues in each group were stained with HE to observe the rejective activity index (RAI) under light microscope. The survival time of the mice was observed.
Results
2
The ratios of activated KCs after liver transplantation at 24
48 and 72 h were 15.9%
21.6% and 28.3%
respectively
and the protein level of TIM-4 in liver tissues was significantly higher than that in the sham group in 1
3 and 7 d after operation (
P
<0.05). The AI (29.23±2.56) of the control mAb group was significantly higher than that (0.40±0.49) in sham group
and that (11.04±2.28) of TIM-4 mAb group (
P
<0.05). Then
after KCs and naive CD4
+
T cells were co-cultured
the growth ratios of CD4
+
T cells were 32.5%
31.6% and 17.2%
respectively in the control
control mAb and TIM-4 mAb groups
and the differentiation ratios of CD4
+
CD25
+
Foxp3
+
T cells were 12.4%
13.9%
and 28.1%
respectively. The IL-4
IL-6 and IL-13 levels in TIM-4 mAb group were significantly lower than those in control mAb group (
P
<0.05). However
the addition of TIM-4 antibody blocked the increased expression of p-STAT6 in CD4
+
T cells co-cultured with TIM-4
+
KCs but not in the cells co-cultured with TIM-4- KCs (
P
<0.05). Furthermore
exogenous IL-4 was added to the co-culture model and overwhelmed the inhibition of p-STAT6 expression by TIM-4 mAb (
P
<0.05). After injection of iTreg cells through the portal vein
the index of liver function in the iTreg group was improved compared with the LT group (
P
<0.05). And the mean RAI score in the iTreg group was 3.97±0.67
which was significant lower than 8.47±0.90 in the LT group (
P
<0.05). The survival time of the iTreg group was markedly prolonged.
Conclusion
2
The blockade of TIM-4 function in Kupffer cells can inhibit the activation of IL-4/STAT6 signaling and induce proliferation of iTreg cells
and as a result
better immune tolerance is induced after liver transplantation.
