Effect of SOX15 gene on proliferation, apoptosis, migration and invasion of colorectal cancer Caco2 cells
To detect the expression level of SOX15 gene in colorectal cancer cells and its effect on cell proliferation
apoptosis
migration and invasion.
Methods
2
The expression of SOX15 gene in colorectal cancer cell lines Caco2
SW480
HCT116
and HT29 and normal colon tissues was detected by RT-PCR. The recombinant plasmid pEGFP-N1-SOX15 and empty vector pEGFP-N1 were transfected into Caco2 cells by liposome Lipofectamine 2000
and the mRNA and protein expression levels of SOX15 gene in Caco2 cells were detected by RT-PCR and Western blotting
respectively. The viability of Caco2 cells transfected with SOX15 was tested by CCK-8 kit
and the cell proliferation was observed by clone formation assay. The regulatory effect of SOX15 on the cell migration and invasion was analyzed by Transwell assay. Flow cytometry was applied to analyze the cell apoptosis.
Results
2
The mRNA and protein levels of SOX15 in the Caco2
SW480
HT29
and HCT116 cells were significantly lower than those in the normal tissues. The mRNA (1.23±0.10) and protein (1.13±0.08) levels of SOX15 in the pEGFP-N1-SOX15 transfected group were obviously higher than those in the pEGFP-N1 control group (0.54±0.06 and 0.19±0.03) and in the blank control group (0.51±0.03 and 0.14±0.02
P
<0.01). Compared with the blank control group and the pEGFP-N1 control group
the pEGFP-N1-SOX15 transfected group was evidently inhibited in cell viability
proliferation
migration and invasion abilities (
P
<0.01)
but cell apoptosis was induced (
P
<0.01).
Conclusion
2
SOX15 gene shows low expression in colorectal cancer cells. SOX15 over-expression can noticeably inhibit the proliferation
migration and invasion abilities
and promote cell apoptosis in Caco2 colorectal cancer cells.
