OBJECTIVE: To observe the therapeutic effects of terdizolamide on methicillin-resistant Staphylococcus epidermidis( MRSE) implant associated osteomyelitis by establishing a rat model of MRSE associated osteomyelitis.METHODS: The stainless-steel wire prefixed with MRSE biofilm was implanted into the proximal tibia of rats. One week after infection
94 rats were randomly divided into the control group( n = 18)
terdizolamide treatment group( n = 16)
terdizolamide combined with rifampicin treatment group( n = 15)
rifampicin treatment group( n = 15)
vancomycin combined with rifampicin treatment group( n = 15) and vancomycin treatment group( n = 15). One week after modeling infection and after completion of treatment
quantitative bacterial culture of bone and steel wire and HE staining of bone tissues of 5 rats in each group were performed respectively. RESULTS: One week after infection
the median number of Staphylococcus cultured on tibia was 4. 89 lg CFU/g [interquartile range( from 3. 83 to 5. 33) lg CFU/g ]
and Staphylococcus were detected in the implanted steel wires of each group of rats. After three weeks of drug treatment
the median of Staphylococcus cultured on bone was 3. 70 lg CFU/g. Except for vancomycin treatment group( 2. 78 lg CFU/g)
the number of Staphylococcus on bone in all treatment groups were significantly lower than those in the untreated group. No Staphylococcus was detected in the rifampicin treatment group
terdizolamide combined with rifampicin treatment group and vancomycin combined with rifampicin treatment group. CONCLUSIONS: The rat model of MRSE implant associated osteomyelitis is sensitive to rifampicin
terazolamide and vancomycin. Tedizolamide combined with rifampicin has better therapeutic effects in the MRSE exogenous osteomyelitis rat model.
