Study on Transmembrane Transport Mechanism of Novel Oral Nanomicelles
OBJECTIVE: To investigate the transport mechanism of polyethylene glycol-distearoyl phosphatidylethanolamine( PEG-DSPE) micelles in monolayer epithelial cells. METHODS: Coumarin 6 loaded PEG-DSPE micelles were prepared by the thin film hydration method
and the fluorescence analysis method for the determination of coumarin6 content was established. The uptake and adsorption of coumarin 6 micelles by Madin Darby canine kindey( MDCK)cells was investigated
and the effects of temperature and different endocytosis inhibitors on the transmembrane transport of coumarin 6 micelles in MDCK cells were analyzed. RESULTS: Coumarin 6 loaded PEG-DSPE micelles could be quickly adsorbed and taken up by cells
the adsorption and uptake reached a dynamic equilibrium. The transmembrane transport of micelles on monolayer cells was affected by temperature
and the transmembrane transport was significantly inhibited at 4 ℃ . Methyl-β-cyclodextrin
hyperosmolar sucrose
chlorpromazine and nystatin all had significant inhibitory effects on the endocytosis of micelles
and the inhibitory rate of methyl-β-cyclodextrin was nearly 88%.Amiloride had no effect on the endocytosis of micelles. CONCLUSIONS: The transmembrane transport of micelles on MDCK monolayer cells is an active and energy-dependent process
and the mechanism of transmembrane transport is multifactorial
including fossil-mediated endocytosis and clathrin-mediated endocytosis.
