Effects of Dexmedetomidine on Inflammation,Apoptosis and Chemotherapy Sensitivity of Lung Cancer A549 Cells
OBJECTIVE: To investigate the effects of dexmedetomidine on inflammation
apoptosis and chemotherapy sensitivity of lung cancer A549 cells. METHODS: A549 cells group
5-fluorouracil group( 40. 0 μg/ml)
dexmedetomidine group( 80. 0 μg/ml)
combination group( 40. 0 μg/ml of 5-fluorouracil + 80. 0 μg/ml of dexmedetomidine) were set up. The above cells were placed in CO
2
incubator( 37 ℃
5% CO
2
2% O
2
93% N2).Six parallel samples were set in each hole of each group for 72 h. At the end of culture
the levels of interleukin-2( IL-2)
interleukin-6( IL-6)
tumor necrosis factor-α( TNF-α) protein in A549 cells were measured by enzyme linked immunosorbent assay
cell viability and number of monoclonal formation were measured by MTT and crystal violet staining
apoptosis was measured by flow cytometry
invasion and migration was measured by Matrigel membrane
nuclear factor-κB( NF-κB)
c-myc mRNA and protein were measured by quantitative real time fluorescent PCR( qRT-PCR) and Western Blot. RESULTS: Compared with A549 cell group
the levels of IL-2
IL-6
TNF-αprotein
optical delnsity( OD)
survival rate
number of cell clone formation
number of membrane penetration
NF-κB
c-myc mRNA and protein expression in 5-fluorouracil group
dexmedetomidine group and combination group decreased significantly
the apoptosis rate increased significantly
the differences were statistically significant( P
<
0. 05).Compared with 5-fluorouracil group and dexmedetomidine group
the levels of IL-2
IL-6
TNF-α protein
OD
survival rate
number of cell clone formation
number of membrane penetration
NF-κB
c-myc mRNA and protein expression in the combined group decreased significantly
the apoptosis rate increased significantly
the differences were statistically significant( P
<
0. 05). CONCLUSIONS: Dexmedetomidine can inhibit the inflammation
proliferation and invasion of lung cancer A549 cells
promote the apoptosis and increase the chemotherapy effects of 5-fluorouracil on lung cancer A549 cells. The mechanism is related to that dexmedetomidine can enhance the inhibition of 5-fluorouracil on the expression of NF-κB
