Effects of Prescription of Kidney-Tonifying and Pulse-Invigorating on Neurological Function,Blood-Cerebrospinal Fluid Barrier and Hemorheology Indicators in Rats with Acute Cerebral Infarction Ischemia Reperfusion
OBJECTIVE: To probe into the effects of prescription of kidney-tonifying and pulse-invigorating on neurological function
blood-cerebrospinal fluid barrier and hemorheology indicators in rats with acute cerebral infarction ischemia reperfusion. METHODS: 180 healthy rats were selected and randomly divided into sham-operation group
model group and prescription group (low-
medium-and high-dose)
with 36 rats in each group. Rat model of reperfusion after middle cerebral artery occlusion were prepared
and successfully modeled rats were selected
with 32 rats in each group. The sham-operation group was given only vascular isolation during the modeling process
without arterial embolization. The prescription group was given prescription of kidney-tonifying and pulse-invigorating for gavage (low-dose group: 25 mg/kg;medium-dose group: 50 mg/kg; high-dose group: 100 mg/kg)
sham-operation group and model group were given an equal volume of 0. 9% sodium chloride solution by gavage for 1 week. The neurological deficits of the rats were detected on the 7th and 14th day of treatment; Evans blue (EB) dye was used to detect the permeability of blood-cerebrospinal fluid barrier;the rat brain tissue was stripped to detect its water content and infarct volume; the kit was used to detect the expression levels of superoxide dismutase (SOD) and malondialdehyde (MDA) in rats; before the rats were decapitated and the brains were taken
the aortic blood was taken to detect the levels of various indicators of hemorheology. RESULTS: On the 7th and14th day of treatment
the neurological deficit score of model group was significantly higher than that of sham-operation group
and that of the prescription group was significantly lower than the model group
with statistically significant differences (P < 0. 05). The EB content
water content
infarct volume and MDA expression level of model group were significantly higher than those of sham-operation group
and those of the prescription group was significantly lower than the model group
with statistically significant differences (P<0. 05). The SOD level of model group was significantly lower than that of sham-operation group
and that of the prescription group was significantly higher than the model group
with statistically significant differences (P<0. 05). The red blood cell deformation indicator of model group was significantly lower than that of sham-operation group
the rigidity index
aggregation index
sedimentation and hematocrit of red blood cell were significantly higher than those of sham-operation group; the red blood cell deformation indicators of rats in medium-dose and high-dose prescription group were significantly higher than that of the model group
the rigidity index
aggregation index
sedimentation and hematocrit of red blood cell were significantly lower than those of the model group
with statistically significant differences (P < 0. 05). CONCLUSIONS: Prescription of kidney-tonifying and pulse-invigorating can effectively repair neurological function in rats with acute cerebral infarction ischemia reperfusion
improve various hemorheology indicators
which has a certain protection on blood-cerebrospinal fluid barrier.
