Research on Dihydrotanshinone Ⅰ Inhibiting Ovarian Cancer Progression Through Nuclear Factor Kappa B Signaling Pathway
OBJECTIVE: To probe into the effect of dihydrotanshinoneⅠon biological behavior of ovarian cancer cell based on nuclear factor kappa B( NF-κB) signaling pathway. METHODS: Tetrazolium colorimetry
cell scratch test
flow cytometry and Western blot were adopted to detect the effect of different concentrations of dihydrotanshinone Ⅰ on the proliferation
migration and apoptosis of ovarian cancer cell strain SKOV3 and the protein expression of NF-κB signaling pathway. RESULTS: Compared with the concentration of 0 μmol/L
the optical density( OD) of SKOV3 cell was significantly lower under the processing of 2
4
6
8 and 10 μmol/L dihydrotanshinoneⅠ
with significantly longer scratch length and higher apoptosis rate; compared with the application of dihydrotanshinoneⅠ for 24 h
the OD of SKOV3 cell was significantly lower after 48 h
with significantly longer scratch length and higher apoptosis rate
with statistically significant differences( P<0. 05); with the increase of the concentration of dihydrotanshinone Ⅰ
the inhibition of proliferation and migration of SKOV3 cells by dihydrotanshinoneⅠand the promotion of apoptosis of SKOV3 cells are more obvious
which are dose-dependent and time-dependent. The comparison of relative expression among NF-κB
P65 and B-cell lymphoma-2( Bcl-2) protein of SKOV3 cells after the processing of different concentrations of dihydrotanshinone Ⅰwere statistically significant( P<0. 05); with the increase of concentration of dihydrotanshinoneⅠ
it had more obvious inhibiting effect on the expression of NF-κB
P65 and Bcl-2 protein of SKOV3 cells
with dose-dependent. CONCLUSIONS: DihydrotanshinoneⅠmay inhibit the proliferation and migration of human ovarian cancer cell line SKOV3 by down regulating the expression of NF-κB
P65 and Bcl-2 protein
inhibiting the NF-κB signaling pathway and inducing apoptosis.
